Ziele, Inhalte und Methode der Lehrveranstaltung

The course provides a comprehensive overview of strategies and methods for protein expression and purification and highlights the utility of different methods of protein characterization for understanding its structure and function. Course graduates are expected to understand and be able to explain theoretical background and instrumentation used for protein characterization to the extent sufficient for critical interpretation of original research results and be able to choose an adequate method for a specific research question. Specifically, the course includes lectures on the fundamentals of recombinant protein expression, purification, biophysical characterization and quality assessment, static and dynamic light scattering, X-ray crystallography, absorption and fluorescence spectroscopy techniques (CD, FRET, FLIM, FC(C)S, fluorescence anisotropy, etc), mass spectroscopy and imaging methods (light and electron microscopy). The course is a lecture series (Ringvorlesung) with elements of flipped class. Additional consultation hours will available using online live conference tools.

Art der Leistungskontrolle und erlaubte Hilfsmittel

Assessment is based on the results of the written exam (120 min on-site or 130 min online, including download/upload time) at the end of the lecture series allowing participants to gain a maximum of 100 percentage points. COVID19 rules: Depending on the epidemiological situation and government and university rules at the time of the exam, the exams will be held either on site or online (via Moodle). Both on-site and online exams will be held as open-book, with textbooks of your choice and lecture notes available as allowed materials. All other materials and all forms of help from third persons are not allowed. The sources of direct and indirect quotes must be properly referenced. Plausibility tests could be held within 4 weeks (28 days) after each online exam.
Active participation in the course (documented through attendance of ALL live online consultation sessions AND posting of at least 8 (eight) original topic-related questions per participant in the course Moodle forum before the 1st exam date) will be counted as a maximal of 15% bonus points to the final exam.

To attain the pass grade, more than 50% points are necessary. Grading scheme: 1 (excellent): 85-100%; 2 (good): 75-84.99%; 3 (satisfactory): 65-74.99%; 4 (sufficient): 50-64.99%; 5 (not sufficient): <50%

Basic principles of absorption and emission (fluorescence) spectroscopy (quantum yield, excitation/emission spectra, Stokes shift, Lambert-Baer's law, CD spectroscopy, etc)
Fluorescence correlation spectroscopy
Basic principles of NMR spectroscopy
Basic principles of mass spectrometry
Basic principles of digital light microscopy (contrast, resolution, magnification, electronic detection of light, binning, factors determining FRET efficiency, etc)
