Aims, contents and method of the course

In this laboratory course, prokaryotes in environmental and medical samples will be detected and identified using the full cycle rRNA approach and FISH with rRNA-targeted oligonucleotide probes. The experiments performed will provide in-depth insights into parameters relevant for nucleic acid hybridization techniques and will expose students to epifluorescence- and confocal laser scanning microscopy. In addition, attendants will learn how to apply digital image analysis to extract quantitative data from FISH experiments.

Assessment and permitted materials

Continuous assessment of course work. Cooperation during the course, written protocol, seminar presentation, written exam.

Minimum requirements and assessment criteria

Acquisition of methods to extract DNA from environmental samples, to PCR-amplify and clone specific genes, and of FISH and fluorescence microscopy. Use of these methods on participants¿ own environmental samples and evaluation of the results.

Examination topics

Molecular biological methods (DNA extraction, PCR, cloning, FISH), microscopy, digital image analysis.
