Ultradünnschnitten; Einführung. in die Tokuyasu-Methode zur Herstellung von Kryoschnitten für Immunogoldmarkierungen; Postembedding-Immunogoldmarkierung vonAcryl-Semidünnschnitten; Evaluierung der Präparation u. Diskussion der Resultate am Fluoreszenzmikroskop, Transmissions-u. Rasterelektronenmikroskop.
Art der Leistungskontrolle und erlaubte Hilfsmittel
NOTE: Students will give a 5 minute ‚Topic of Interest’ introduction talk including a description of the samples they will use in the course. This talk will be immediately followed by a 5 minute discussion for recommendations on preparation of their samples for the course.All students are kindly requested to provide an abstract (max. 200 words) describing your major research interest and the intended application of immuno-electron microscopy in your research. This abstract must be received by email to Dr. Jacqueline Montanaro and Dr. Siegfried Reipert. Latest acceptance of abstract is 14. October, 2013.; ; In addition, please provide the following information:; Your Name, Institute, Department, Phone number, email, webpage and please indicate (yes or no) whether you have previous experience with ; LRW embedding, TEM, Epifluorescence microscopy, ultra cryo-microtomy and any immuno-staining techniques at light microscopy or electron microscopy level.; ; Please send abstracts and further information to:; jacqueline.montanaro@meduniwien.ac.at and siegfried.reipert@univie.ac.at. ; ; You are advised that you will be required to give a 5 minute introductory talk during the course (04.10.13) describing your major research interest and the intended application of immuno- electron microscopy in your research. This is intended to be an introduction of the students to the lecturers and to eachother. Please note; This student introduction talk will contribute towards the final assessment.Student assessment is divided into 4 parts:(1) Introductory Student talk (5 mins) and abstract,
(2) Participation in lectures, group poster preparation and practicum,
(3) Individual oral assessment of understanding technical and theoretical knowledge presented in lectures and demonstrations (end of course),
(4) Final student talk (7-10 mins) followed by open discussion and questions.

Mindestanforderungen und Beurteilungsmaßstab
To enable advanced BIO-stud. to understand & apply advan.cryo preparation techniques to electron microscopy (EM), using immunolabelling within their own research. Supported by lectures & demonst., this intensive, hand-on training course teaches special. techn., such as Tokuyasu cryosectioning, high-pressure freezing, freeze substitution & acrylic resin embedding for high resol. localization of intracell. proteins & other intracell. struc. by immunogold labelling. Plunge freezing and freeze drying for scanning EM will be demonstrated.
