Information
Aims, contents and method of the course
The proteome of a complex sample shall be analyzed. Therefore, first, the complexity of the sample will be reduced using appropriate separation techniques. After that, an analysis of the sample, applyig mass spectrometry, will be performed. In the course of this practical work, different methods for protein analysis will be presented and practically applied.
Assessment and permitted materials
Performance is monitored during the practical course (interest, cooperation), and furthermore determined on the basis of the quality of the delivered protocol and the acquired knowledge (determined in the closing meeting).
Minimum requirements and assessment criteria
Acquisition of the most important knowledge and skills necessary to work with complex biological samples and allow a subsequent proteome analysis using LC-MS/MS.
Examination topics
Background knowledge on the conducted experiments: preparation of biological samples (blood samples), isolation of different blood constitutents (blood plasma and white blood cells), subcellular fractionation (separation of cytoplasmic and nuclear proteins), protein separation techniques using 2D- and 1D-gel electrophoresis, digestion of proteins into petides, identification of proteins using LC-MS/MS analyses.
