Aims, contents and method of the course

Prinziples of protein-ligand interactions (bindung contributions, thermodynamics and binding energetics), principal experimental protocols for the determination of bindung constants (steady-state measurements, kinetic measurements, competition experiments), linear transformations (Scatchard plot), stoichiometry, allosteric effects, cooperativity;
methods for the determination of binding constants: microcalorimetry, radioreceptor assay, chromatographic methods (including static (batch) method for the determination of adsorption isotherms, frontal analysis, Hummel-Dreyer Method), elektrophoretic methods (affinity CE), spectroscopic methods (NMR, UV, fluorescence, CD), surface-plasmon resonance

Minimum requirements and assessment criteria

The goal is that the students get knowledge of the various possibilities for the determination of binding constants by distinct techniques. The course should teach students about the general principles, technical aspects, potential sources for errors and the applicability range of the different techniques.
