Aims, contents and method of the course

The aim of the course is to introduce you to the range of different examination and documentation possibilities in light microscopy and to introduce you to the application here at CIUS. It is primarily intended for those students who will use light microscopy methods in their own research and want to achieve "high end" quality. Techniques in optaining quantitative data from microscopic images will be discussed and applied.
The theoretical content will be presented in the first unit and will be partly developed independently, using literature and video lessons:
- Structure of a microscope and the individual components: Objective types, beam path, resolution & illumination.
- Presentation of various contrast methods (bright field, dark field, phase contrast, DIC, polarization, fluorescence).
The written examination concludes this part and is intended to ensure that knowledge can be deepened in the practical part.
In the subsequent practical part, participants gain an overview of the techniques and in particular the possibilities for measurement and documentation on the light microscopes in the UBB Core Facility (stereo, transmitted light, fluorescence and laser scanning microscopy). You will work on samples in small groups with all types of microscopes and communicate the results to all participants in a PowerPoint presentation to reinforce the memory effect.
As a result of the course, you should be able to select the correct and most efficient method of light microscopy for your own sample and research question.

Assessment and permitted materials

Attendance is compulsory, a maximum of 1 day can be missed due to illness.
The preliminary meeting is considered to be the first teaching unit and is therefore subject to compulsory attendance! If registered students are absent from the preliminary meeting without prior agreement (email!), the course instructor will cancel the course.
Since the preliminary meeting is considered the first course unit and thus course participation has begun, deregistration after the deregistration deadline (15.03.2024) is only possible in justified exceptional cases (e.g. doctor's confirmation). If no sufficiently substantiated justification is provided, this exercise must be assessed negatively.
A single excused absence (8 hours) during the exercise weeks is permitted. Excusable reasons are, for example, a doctor's appointment with confirmation or compulsory attendance at other courses or examinations.
The performance assessment consists of three parts
- Written examination on the theoretical part (weighting 40%)
- Active participation during the hands-on internship (weighting 20%)
- Final presentation on the last day of the course or on a day agreed in the preliminary discussion (weighting 40%)
For work in the microscopy laboratory in the Core Facility (CIUS), the internal safety course must be completed, this counts towards the total hours. It is mandatory to bring a lab coat on the days when samples are prepared. It is assumed that previous experience with course microscopes has already been gained in previous practical courses and that laboratory experience and computer skills are available.

Minimum requirements and assessment criteria

The course is assessed positively (grade 4) if 55% of the possible points are achieved in both the written examination and the final presentation (15 min speaking time for the group + 5 min discussion). Each partial performance is assessed separately and is included in the overall assessment of the course in different percentages (see above).
