Die Übung "Molekulare Genetik und Pathologie" ist an einen positiven Abschluss der Vorlesung "Zellbiologie tierischer Zellen" (Mikulits, Dolznig, Müllner) (SS) gebunden.
Die Übung verläuft gemeinsam mit dem "Seminar in Zellbiologie". Das "Seminar in Zellbiologie" kann alleine nicht absolviert werden.

Aims, contents and method of the course

The practical course deals with 2-dimensional (2D) and 3-dimensional (3D) cell cultures of human carcinoma cells and carcinoma-associated fibroblasts. Experiments focus on the analysis of gene expression after RNA interference (RNAi), examination of the impact of conventional and targeted therapy on cell proliferation and clonogenic growth behavior in 2D and 3D cultures after generation of microspheres. In addition, the practical course concentrates on the analysis of hematopoiesis by flow cytometry, on the histology of various tissues and immunohistochemical analyses of experimental tumors after chemical induction and xenotransplantation of carcinoma cells.

Evaluation of the experimental work and quality of the protocol.

Minimum requirements and assessment criteria

The students develop deeper and more comprehensive knowledge about the pathophysiology of tumorigenesis and its determinants. The students acquire technical skills and get access to the experimental methods of molecular cell biology to study the mechanisms of tumorigenesis.

Examination topics

2D and 3D cell cultures, generation of microspheres, RNAi/small interfering RNA, analysis of gene expression, invasion of spheres in collagen gels, treatment of 2D and 3D cell cultures with conventional and targeted chemotherapeutics, analysis of cell proliferation, clonogenicity and apoptosis after administration of chemotherapeutics, determination of cellular senescence, analysis of cell migration by woundhealing/scratch und Transwell assays, synchronization of the cell cycle, performance and interpretation of flow cytometry, histology and immunhistochemistry.
