Aims, contents and method of the course

Students become acquainted with varied preparation-techniques of living plant and animal material for study with light-, scanning electron-, and transmission electron microscopy (LM, SEM, TEM) in both theory and practice. Students prepare specimens via fixation, embedding, and staining and document their results using different microscopes. These methods are applicable for diverse organisms (plant and animal tissues) and therefore relevant for different disciplines of biology. Students will learn how to apply appropriate methods in order to answer future research questions within their field of biology.
• Microtomy (sectioning, 5 µm thin)
• Fixation (application of different methods and comparison)
• Staining techniques (toluidine blue/ruthenium red, acetolysis)
• Light microscopy (LM)
• Scanning electron microscopy (SEM)
• Transmission electron microscopy (TEM)

Assessment and permitted materials

Based on a written protocol (60%) and active participation (40%) throughout the course.

Minimum requirements and assessment criteria

Students prepare specimens for LM (microwave fixation, Kulzer-embedding, microtomy, different staining techniques) and SEM/TEM, they document specimens digitally, analyse, and interpret their own results.

Examination topics

Lectures, demonstrations, and practical work throughout the course.
