Aims, contents and method of the course

Our "Live Imaging Course" was markedly re-scheduled based on the evaluation process of teachers and students: The course is planned for three days and 12 attendees will be allowed. Newly integrated is a superresolution microscopy experience, as well as a session dedicated on handling and processing data from live imaging experiments.
The Live Imaging workshop intends to give students a theoretical and practical input on important considerations of live imaging using advanced light microscopic technologies, like confocal, spinning disc, fluorescence widefield and even superresolution imaging microscopy.
In total now 7 lectures on individual topics will be held:
- Live Imaging Considerations
- Sample preparation
- Single molecule superresolution microscopy
- theoretical comparison of course microscope modalities
- Dyes and Fluorophores
- Specialized Techniques (FRAP, AIRY SCAN, LATTICE SIM)
- Handling Live Imaging Data

The 12 attendees will become split into three groups of 4 people. Each group will work on one of the three offered microscopy modalities (widefield, confocal, spinning disc) throughout the course.
Extensive hands-on planning of overnight and individual shorter time lapse experiments using various available microscope setups will be considered. Special applications at each modality, such as photomanipulation, superresolution options and/or specialized processing are part of the training.
Two groups of 6 people will have the chance to get a home-built superresolution microscope setup demoed.
The final afternoon is dedicated to give expert advice on how to handle live imaging data - this will include selected examples as well as insight into advanced processing pipelines, including aspects of Artificial Intelligence.

Minimum requirements and assessment criteria

Students shall learn the theoretical background of imaging in a live regimen. This includes knowledge on instrumental and technological theory, combined with considerations on practical do's and don'ts. The latter shall finally be applied in extended practical hands-on-sessions using different live microscopy setups. Eminently important is to prepare students for handling the data with respect to size, processing and analyses.

Examination topics

a. Students will understand the theory behind basic and advanced light microscopy technologies
b. Students will get a deeper insight into the applicative potential of diverse light microscopy techniques for live imaging
c. Students will learn to transfer knowledge on microscopy hardware and fluorescent molecule selection into an experimental realization
d. Students will learn to critically review, troubleshoot and/or optimize live imaging experiments
e. Students will learn and work in cooperative teams
f. Students will get a comprehensive picture by being introduced into data management
g. a quiz will help evaluate the learning success, as questions are to be answered before the course starts and also at the end of the course
