Information

Aims, contents and method of the course

The course communicates theoretical and practical knowledge necessary to quantify nucleic acid sequences using reverse-transcription quantitative PCR (RT-qPCR), but also digital PCR, the current gold standard for precise counting of (low) DNA copy numbers. Examples from the fields of microbiology and immunology will be addressed. Major experimental topics are the use of reference sequences like spike-in controls for molecular diagnostics, normalizers required to determine relative transcript expression and counting of absolute numbers of DNA copies. Practical training includes primer design for monitoring of targets like mRNA variants, micro RNAs and DNAs by a fluorescent dye or various probe formats. The course also aims at developing basic skills and abilities in statistical evaluation and graphical presentation of the data, as well as trains storytelling in written communication.
The course is designed for master students of the final trimester. Maximum of participants: 15

Evaluation consists of different performances:
• Answers in an initial questionnaire based on information provided by the review article: Kralik P, & Ricchi M, 2017 A Basic Guide to Real Time PCR in Microbial Diagnostics: Definitions, Parameters, and Everything. Frontiers of Microbiology 8: 108),
• Active participation and results obtained,
• Theoretical knowledge including oral presentation of a methods article,
• Colloquium on assay design and data evaluation,
• One-page protocol report (scientific article about one of the data sets or assay designs)

Minimum requirements and assessment criteria

Compulsory attendance; 100 points can be reached:
• Answers of introductory questionnaire: 20 points (20%)
• Active participation: 20 points (20%)
• Presentation of a methodological article: 20 points (20%)
• Protocol (scientific article on one page): 40 points (40%)
All subgrades must have a positive evaluation.

Examination topics

Review article on quantitative PCR (see literature), content of the presentation and the protocol on one experimental data set or assay design.
