Aims, contents and method of the course

The following topics will be covered the major topic of macromolecular crystallography and will provide students with a comprehensive overview of all the steps required to generate a three-dimensional structural information on biological macromolecules:
* sources of X-rays
* crystallisation techniques
* symmetry (point groups, space groups), unit cell, Bravais lattice
* scattering of X-rays, atomic form factor, structure factor
* waves, diffraction of molecules and crystals, Fourier transform; electron density
* set-up of MX diffraction experiment and detectors
* methods for solution of phase problem: molecular replacement, multiple isomorphous replacement, multi-wavelength anomalous dispersion
* density modification techniques
* structure refinement, structure validation and analysis

Assessment and permitted materials

Oral or written exam, depending on the number of students.
Oral exam in Zoom or written digital exam.

Minimum requirements and assessment criteria

Attendance and active participation during the lecture course, >60% correct answers of exam for passing grade.
Assessment criteria:
100-90%: excellent (1)
89-80%: good (2)
79-70%: satisfactory (3)
69-60%: sufficient (4)
< 59%: insufficient (5)
