Objective: The objective of the course is to teach the student in experimental techniques used in organic synthesis. This is illustrated by a number of important organic reactions. Practical training is given in this laboratory course.
Course Description: Glassware of a preparativ organic laboratory; weighting; adding of compounds to a reaction vessel; control of temperature; extraction techniques; distillation (at atmospheric pressure - at diminished pressure); fractional distillation; recrystallation; drying of solids and liquids; determination of melting points; steam distillation; azeotropic distillation - water separation unit, adding of compounds via septum and syringe under a nitrogen atmosphere; recording of results; general instructions for safe working.
Program: Separation of a mixture of two liquid compound by fractionated distillation under atmospheric pressure and diminished pressure; purification of a contaminated solid compound; isolation of an org. acid or base, respectively, by extraction.
Eight syntheses: Reactions of carboxylic derivatives (esterification, acetylation); reactions of carbonyl compounds (acetals-, imines-, enamines- und oximes); condensation and addition ractions and rearrangement reaction (Aldol-, Knoevenagel-, Mannich-, Perkin-, Diels-Alder-reactions, benzilic acid rearrangement reaction); reductions (komplex hydrides) and oxidation reactions; syntheses of heterocyclic and aromatic compounds; Grignard-reactions (use of septum and syringe; nitrogen atmosphere); preparation of drugs.
Preconditions: Successful completion of the laboratory course "Quantitative pharmaceutical analysis" and of the lecture "Drug Synthesis - Basic Concepts (incl. nomenclature)".
Examination: Evaluation is based on laboratory work.
