321023 VO Methods of Separation and Analysis of Org. Drugs incl. Bioanalytical Methods - BA12 (2016S)

The first part of the lecture deals with pharmacy-lab relevant methods for the analysis of drug compounds (APIs) and with the pharmaceutically relevant separation methods, especially for small molecules. In the second part of the lecture, established methods for the analysis of (pharmaceutically relevant) biomolecules are treated, with a special focus on proteins and nucleic acids.

Aims of part 1: providing basic knowledge and understanding of qualitative analytics of drug compounds with methods that can be applied in a community pharmacy (identity check of APIs); knowledge about the underlying priciples, scope and limitations of separation techniques used in instrumetal abalytics (focus: chromatographic methods).

Aims of part 2: providing basic knowledge of analytical methods used in the bio sciences. These methods are essential both in the production and quality assurance of pharmaceutical products known as “biologicals” (proteins, glycoproteins, drugs produced with the aid of recombinant DNA,…) as well as in the diagnosis of diseases (genomics, DNA chips).

Contents of part 1: determination of the elemental composition, solubility, precipitation reactions, colour reactions, proof of functional groups in drug molecules, physicochemical properties (melting point, refraction index); chromatographic methods (underlying mechanism, classification, theoretical principles); different experiemntal thechniques: thin-layer chromatography (TLC), column chromatography (CC), high-performance liquid chromatography (HPLC), gas chromatography (GC); discussion of their characteristic features, necessary equipment, development techniques, detection options, sample preparation, derivatisation; examples of pharmaceutical applications; determination of the separation performance of a chromatographic system; quantitative aspects in chromatography, working with internal and external standards; validation of methods

Contents of part 2: I. Peptides and proteins: Basics, synthesis of peptides, purification of proteins, electrophoresis, methods of structure determination, quantification, chemical modification, radioactive labeling of proteins; proteomics; glycoproteins.
II. Nucleic acids: Basics, synthesis of oligonucleotides, purification of nucleic acids, analytical methods (hybridization, DNA sequencing, DNA chips).
III. Enzymatic analysis
IV. Biosensors
