Aims, contents and method of the course

During the practical course basic molecular biology techniques will be applied:
" Isolation of leucocytes from blood samples and DNA extraction
" Primer design for amplification of the region of interest of the cytochrome P450 2D6 gene
" Amplification of the region of interest by polymerase chain reaction
" Purification of the amplified DNA fragment and DNA digestion with restriction endonucleases
" DNA separation by agarose gel electrophoresis and gel documentation

Minimum requirements and assessment criteria

This course is intended to provide insights into the methodology of pharmacogenetics.
The pharmacokinetics of many drugs is affected by activity of specific cytochrom-P450-enzymes. Genotyping of test subjects (the course participants) will be performed by analysing genomic DNA. The polymorphism of cytochrom-P450 2D6 concerning a particular mutation will be determined. The therapeutic relevance of Cytochrome P450 polymorphism will be discussed.
